Effect of Jianpi Yiqi Koufuye on small intestine propulsion in mice and analysis of its mechanism

To investigate the effects of Jianpi Yiqi Koufuye on functional dyspepsia and its mechanism of action. Sixty KM mice, half male and half female, were randomly divided into blank group, model group, and Jianpi Yiqi Koufuye (high, medium, low, and very low dose) group, administered orally for 4 weeks, and determined by the ink method small bowel advancing rate. Screen disease targets through GeneCards and Drugbank databases. Search BATMAN-TCM for the chemical constituents and their targets in Chinese herbal medicines. Through the String database and Cytoscape software, a target interaction network was constructed, key targets were extracted, and GO, KEGG enrichment analysis, and network visualization analysis were performed. Virtually screen compounds and targets related to drug efficacy through Discovery Studio and analyze their binding patterns. The Jianpi Yiqi Koufuye group significantly improved the small bowel advancing rate of the mice compared with the model group (P<0.01). A total of 194 chemical constituents and 703 chemical constituent targets were screened from the prescription. There were 601 targets for functional dyspepsia.A key network consisting of 104 targets, including TNF, INS and MAPK1, was extracted from the target network, which was involved in the regulation of the secretion of peptide hormones, protein phosphorylation and other biological processes, as well as the regulation of TNF signaling pathway, IL-17 signaling pathway, cAMP signaling pathway, cGMP-PKG and Ca2+ signaling pathway. Molecular docking results showed that in 194 chemical components, niacin and phenylalanine contained in codonopsis codonopsis and chicken gold bound with FOS, EGFR, CREB1 and NOS3 proteins in Ca2+ signaling pathway and cAMP signaling pathway by hydrogen bond and van der Waals force. Jianpi Yiqi Koufuye promotes the small intestine's advancing function in mice, and its mechanism may be related to the expression of FOS, EGFR, CREB1, and NOS3 in the cAMP signaling pathway and Ca2+ signaling pathway, which are involved in the regulation of nicotinic acid and phenylalanine. Jianpi Yiqi Koufuye can be used to treat functional indigestion.
