In order to study the effect of melatonin on the salt resistance of tartary buckwheat, the seeds of an inbred line WTX2 of Tartary buckwheat were soaked with different concentrations of melatonin and water as control (CK). Then the germination experiment was carried out under salt stress (100 mmol/L NaCl). The results showed that the salt resistance of seeds treated with melatonin was significantly improved compared with CK. Seed vigor index and seedling fresh weight treated by 100 and 1 000 μmol/L melatonin were higher than those of CK. The malonaldehyde (MDA) content in seedlings treated with 100 μmol/L and 1 000 μmol/L melatonin increased slowly during germination under salt stress, while it increased sharply in CK seedlings on the 6th day of germination. The seeds treated with 100 and 1 000 μmol/L melatonin had no significant decrease in superoxide dismutase (SOD) activity during 4~6 days of germination, while the SOD activity in CK seedlings significantly decreased. There was no significant change in catalase (CAT) activity of seedlings treated with 1 000 μmol/L melatonin germinated under salt stress, while the activity of CK seedlings showed an obvious decreasing trend. Appropriate concentration of melatonin can effectively alleviate the stress of high salt on tartary buckwheat.

To investigate the effects of different nitrogen application rates on yield and agronomic characteristics of cultivar-Taoyouxiangzhan under the rice-crayfish co-culture, 5 nitrogen application rates 0 kg/hm2(N0), 135 kg/hm2(N1), 150 kg/hm2(N2), 165 kg/hm2(N3), 180 kg/hm2(N4) were conducted in this study. The results showed that the maximum tiller number increased by 30%, the SPAD value of leaves increased by 9.38%, and the above-ground dry matter of the spikes at heading stage and milking stage increased respectively by 37.9% and 41.1%. The effective spikes, seed setting rate and thousand-grain weight were respectively increased by 39.1%, 16.3% and 3.69%. The actual yields under the conditions of N1, N2, N3 and N4 were respectively increased by 57.8%, 54.1%, 69.2% and 61.6% compared to the control. The effect of increasing yield is best under the condition of N3, but excessive nitrogen fertilizer application reduces nitrogen fertilizer utilization rate and causes agricultural non-point source pollution. Therefore, rational application of nitrogen fertilizer is of great significance for improving rice yield and protecting the environment.

The comprehensive zoning of ecological restoration difficulty of wind farms in Hubei province was carried out on the basis of full consideration of meteorological effects, which provided theoretical basis for wind farms in different regions to take different ecological restoration measures. 12 index factors of five categories were selected, including meteorology, soil, vegetation, geographic information and wind energy resources, which affected the ecological restoration of wind farms in Hubei province. The meteorological factor model was established in different regions by using the observation data of 89 national meteorological observation stations and 2 005 automatic observation stations from 2016 to 2018, and the distribution of annual rainfall, summer rainfall, annual mean minimum temperature and other indicators were obtained. A simplified soil rainfall erosivity model was established by using monthly precipitation, and the soil rainfall erosivity index was obtained. The land use type index was obtained by using land cover classification data. The normalized vegetation index (NDVI) was obtained by using satellite remote sensing data. Through the high-resolution (1 km) assessment (30-year average) data set of land wind energy resources, the wind speed at 70 m height and the wind power density at 70 m height were obtained. On this basis, based on the idea that meteorological impact played a dominant role in the ecological restoration process of wind farms, different weight influence coefficients were given to each index factor. Through the weighted comprehensive score method, the comprehensive regionalization grades of wind farm ecological restoration were obtained, which were easy recovered areas, easier recovered areas, difficult recovered areas and more difficult recovered areas. The easy recovered areas were mainly distributed in Jianghan plain, hilly plain in the middle of Xiangyang, Xishui, Qichun and Tuanfeng in east of Hubei province; The easier recovered areas were mainly distributed in the hilly areas of southeast and northwest Hubei; The difficult recovered areas were mainly distributed in the Dabie mountain area in the northeast of Hubei province, the Mufu mountain area in the southeast of Hubei province, the Tongbai mountain area in the north of Hubei province, the Dahong mountain area, and most area of Enshi. The more difficult recovered areas were distributed in some middle and high mountain areas of Hubei province.

In order to understand the spatial distribution and influencing factors of total selenium in cultivated land of Lichuan city, Hubei province, the total selenium content of 668 cultivated land samples were determined. The results showed that the measured values of total selenium in cultivated land presented lognormal distribution. The content of total selenium ranged from 0.011 mg/kg to 6.121 mg/kg, and the arithmetic mean and geometric mean were 0.465 and 0.349 mg/kg. In the land area of Lichuan city, the total selenium adequacy rate was 92.0% (more than 0.175 mg/kg). The findings showed that Lichuan was a selenium rich area in China and an important part of the “selenium capital” of Enshi. There were significant differences in total selenium content among different soil parent materials, soil types, land use patterns or altitudes. In most of the factors studied, total selenium content was positively correlated with soil organic matter content and altitude, but not with pH. The content of selenium and organic matter in the soil of each factor (except the utilization mode) changed synchronously. Due to the influence of parent materials, soil type, land use patterns or altitude and other factors, there were differences in the content of soil organic matter, which might lead to changes of total selenium content distribution in cultivated soil of Lichuan city.

In order to quantify the relationship between solar greenhouse temperature and external meteorological conditions under different weather conditions, the weather conditions were analysised based on sunshine percentage method, and the changing rules of temperature in greenhouse under different weather conditions were analyzed. The quantitative relationships between temperature in greenhouse and external meteorological conditions were studied by establishing a regression equation. The results showed that on sunny, cloudy and full-cloudy days, the maximum temperature appeared at 12:00, 12:00 and 13:00 in spring and autumn, and the minimum temperature appeared at 6:00, 7:00 and 7:00 in greenhouse. In winter, the maximum temperature appeared at 13:00, 13:00 and 14:00, and the minimum temperature all appeared at 8:00. The temperature rising time in greenhouse was about 1 h later than outside, and the cooling time varied, with 2~3 hours earlier than outside in spring and autumn, and 1~2 hours earlier in winter. When the number of consecutive overcast days was less than 3 days, for every 1 ℃ decrease in outdside minimum temperature in spring and autumn, the minimum temperature in greenhouse would decrease by about 0.25, 0.32 and 0.32 ℃ on sunny, cloudy, and full-cloudy days, and by about 0.34, 0.42 and 0.37 ℃ in winter. When the number of overcast days exceeds 3 days, for every 1 days of continuous overcast days, the minimum temperature in greenhouse would decrease by 0.33 ℃ in spring and autumn, and by 0.41 ℃ in winter. For every 1 ℃ decrease in the outdside minimum temperature, the minimum temperature in greenhouse would decrease by about 0.34 ℃.

In order to meet the needs of forest production and management, dealing with cases of illegal felling and excessive felling, checking and accepting the felled forest volume, the relationship between the ground diameter and volume of Cryptomeria fortunei was studied and the volume table of Cryptomeria fortunei ground diameter was compiled. According to the resource distribution of Cryptomeria fortunei in Guizhou province, the author uses the typical sampling method, 21 investigation samples were selected from Daozhen, Hezhang, Pu'an, Pingba and Jinsha, and 317 samples wood data were investigated according to 8 diameter steps, among which 240 were constructed and 77 were tested. According to the technical specification for root diameter standing volume table and six mathematical models commonly used for root diameter standing volume. Taking the weighted least square method of ForStat 2.2 statistical software, the best model of Cryptomeria fortunei diameter volume table was selected by comparing and evaluating the fitting effect of 9 indexes such as sum of squares of deviations, correlation index and total relative error. The volume model of Cryptomeria fortunei has some guidance for estimating the volume of Cryptomeria fortunei, but the result is limited because only one variable of ground diameter.Under the condition that the ground diameter, diameter at breast height and tree height are all limited, it is recommended to use the binary standing timber volume model of Cryptomeria fortunei.

Based on the TRU system, the root system of Camellia szechuanensis Chien planted in Nanshan Botanic Garden, Longcha Huahai, and Yongchuan Road and Park Greenbelt was measured at 0.5 m and 1.0 m circle of the trunk, the number and distribution range of diameter ≥1cm roots were measured,in order to compare the distribution characteristics of roots under different site conditions, explore the limit factors of its application in road and park greenbelt. Meanwhile, 7 soil samples were collected, 8 indexes were measured for each soil sample, such as soil pH, EC and organic matter content and so on. The results showed that:(1) at a distance of 0.5 m to 1.0 m from trunk, the depth of diameter ≥1 cm root distribution is increasing, at a range of 0-62 cm; (2) at a distance of 0.5 m to 1.0 m from trunk, the number of diameter ≥1 cm roots increases gradually. Among them, The number of diameter ≥1 cm roots increased from 20 to 35 in Nanshan Botanic Garden, from 34 to 46 in Longcha Huahai, from 11 to 13 in Yongchuan district; (3)78%-85% of diameter ≥ 1 cm of roots distributed in 20-72 soil layer in Nanshan Botanic Garden, 80-82% of diameter ≥1 cm of roots distributed in 0-40 cm soil layer in Longcha Huahai, about 50% of diameter ≥1 cm roots distributed in 0-20 soil layer in Yongchuan district; (4) Compared with Nanshan Botanical Garden and Longcha Huahai, at a distant of 0.5 m and 1.0 m from trunk, the tree planted in Yongchuan district with the same diameter showed less root number, root distribution and root extension. This is closely related to the poor soil quality at the survey site. Anyway, improving the soil quality, especially the depth of main roots distribution, plays an important role in promoting the restoration of C. szechuanensis Chien growth.

Based on the coherence matrix of the image, the characteristic parameters, namely the radar vegetation index (RVI), were extracted. Then the Freeman decomposition of the covariance matrix of the image was performed, and three scattering mechanism parameters were obtained, which are body scattering, surface scattering and dihedral scattering. These parameters are then combined into a support vector machine (SVM) to classify the polarimetric SAR images and compare them with the Wishart supervised classification. The results show that the radar vegetation index can improve the classification accuracy of vegetation, and the classification accuracy of this method was significantly higher than the Wishart supervised classification.

To investigate the effects of six exogenous hormones on the expression of tomato transcription factor SlMYB102 gene under low temperature stress. Exogenous hormone spraying and low temperature treatment were applied to tomato seedlings with four leaves and one heart. In hormone treatment, salicylic acid (SA), augrin (IAA), gibberellin (GA3), 6-benzylamine (6-BA), abscisic acid (ABA) and methyl jasmonate (MeJA) were sprayed on the leaves. Distilled water was sprayed as a control. Sampling at 0, 1, 3, 6, 9, 12 and 24 h after low temperature treatment at 4 ℃. The expression of tomato transcription factor SlMYB102 gene was detected by real-time fluorescence quantitative PCR. The results show that induced by ABA,GA3,IAA,SA,6-BA and MeJA, SlMYB102 gene responded to all six hormones and presented an up-regulated expression pattern. As a member of the R2R3-MYB subfamily of MYB family, SlMYB102 transcription factor may be involved in the regulatory network of these hormones in response to stress.

To identify the influence of Nlu-miRNA-8 on the expression of Ccdc124(coiled-coil domain-containing protein 124-like), based on the construction of the psi-CHECK2 luciferase reporter gene vector. Firstly, the Ccdc124 gene fragment containing the Nlu-miRNA-8 binding sequence was designed and synthesized. The luciferase reporter gene vector was constructed and transfected into Drosophila S2 cells. The effect of Nlu-miRNA-8 on the expression of Ccdc124 was observed by the change of luciferase activity. The results showed that Nlu-miRNA-8 significantly inhibited the fluorescence expression of the Ccdc124 group, but it did not significantly inhibit the fluorescence expression of the blank control and NC group. In this study, we successfully constructed a psi-CHECK2 vector of the 3′-UTR segment of the Cdcc124 gene, and preliminarily confirmed that Nlu-miRNA-8 can regulate the Cdcc124 gene.

The evaluation index system for the implementation of the household garbage classification policy was constructed from six aspects of execution validity, execution strength, execution attitude, execution angle, execution speed and execution difficulty. Then, the AHP method was used to reckon the index weight of each level, and the FCE method was used for comprehensive evaluation. The results indicate that the weight values of the six standard levels are execution strength>execution validity>execution difficulty>execution attitude>execution angle>execution speed. The comprehensive evaluation result of the implementation of the domestic waste classification policy in Jiading district, Shanghai is at a strong level. On the one hand, it shows that there is still room for improvement in the implementation of the domestic waste classification policy in Jiading district. On the other hand, it also shows that the prospects for compulsory classification of domestic waste to be fully spread nationwide are promising. Finally, some suggestions are put forward based on the evaluation results.
