    Gene-specific molecular markers are directly developed from functional SNPs or InDels of the gene itself. It has several advantages such as high selective efficiency, co-dominant, co-separate with the target gene, unaffected by genetic background, high correlation to trait, accuration and directness, exactly track and location of target alleles gene under different genetic background, which is an ideal molecular marker for crop marker-assisted selection breeding. The present review concerned the advances on the principle, development, application and development trend of gene-specific molecular markers, which will provide reference to the molecular assisted breeding.

    Based on the GPS data sounded from GPS/MET networks inn Chenzhou area(Yongxing,Yizhang,Guiyang,Rucheng and Guidong) from January to December 2017,the hourly precipitable water vapor (PWV) has been retrieved by using Saastamoinen hydrostatic delay model and Bevis empirical formula,combining the surface atmospheric pressure and temperature. The accuracy of PWV retrieved from GPS data,the temporal-spatial variation of PWV and the relationship between PWV with other atmospheric elements as well have been discussed. The results indicate that the reliability of PWV retrieved from GPS data is verified compare with the PWV based on the radiosonde observation sub-second data in Chenzhou area,and the mean bias is 1.717 7 mm,the root mean square error is 3.258 0 mm. The monthly mean PWV presented single peak distribution,which reached the global maximum value in summer(June to August) and dropped down to the global minimum value in winter(November to January). Due to surface evaporation and local circulation,the daily variation of PWV presented two peaks distribution that the relative maximum values assumed at about 16:00 and 00:00,and the variation characteristics of PWV could be used as instruction on the precipitation forecast somewhat because the PWV usually increases before heavy rainfall.

    In order to promote the sustainable use of the land in Wuhan Metropolitan Area,to balance the level of land ecological security in all municipalities within the system,explore the status and differences of urban individual and the whole area. Therefore,based on the improved literature reference method and the DPSIR model,the evaluation index system of land ecological security of Wuhan Metropolitan Area was constructed,using principal component analysis to calculate the comprehensive value of urban land ecological security in 2015,then,compared the difference between the whole and the city level. The results show that:the overall ecological security level of Wuhan Metropolitan Area is good,but the development of cities within the group is relatively uneven. There is no direct linear relationship between urban scale,urbanization level and land ecological security level. According to the ecological security value, the nine independent cities that make up the Wuhan Metropolitan Area are divided into four ranks and the corresponding measures are given.

    With the continuous acceleration of the urbanization process, the contradiction between man and land has become increasingly prominent, and the compact urban development mode has become one of the main ways to curb the disorderly spread of the city and alleviate the “big city disease”. According to its core connotation and related theories, this paper takes 13 cities in Changsha, Hunan, Zhuzhou and Xiangtan as the object of study. The compactness evaluation index system is established from three dimensions: high density, reasonable structure and good efficiency. Stepwise regression analysis, the urban compactness and its main influencing factors were analyzed and analyzed to explore the spatial and temporal characteristics of urban compactness in Hunan province. The results show that: the overall level of urban compactness in Hunan province is increasing, the high and medium degree compact cities are increasing, the low degree of compact cities are decreasing, and the gap between cities is gradually widening. The distribution of the high, medium and low degree of compact cities in Hunan province has changed from “water droplets” to inverted “water droplets”, and presents the spatial pattern of rising from west to east. The main factors influencing the urban compactness of Hunan province are economic density, followed by land mixture, population growth elasticity, population density and so on.

    In order to investigate the influence of closed type changes of water quality in shrimp pond environmental factors season on the community structure of planktonic microalgae, test the isolated experiments of pond water temperature,pH,total nitrogen (TN),total phosphorus(TP) and the content of chlorophyll a,and analyse the the same period in water transformation of planktonic microalgae community structure and advantage to explain in foreign sources under the condition of no interference on microalgae community changes in response to environmental factors. The results show:pond water temperature and TP content from April to the end of September increased gradually, declined in October. pH value changed first down, then up. The total nitrogen (TN) and the content of chlorophyll a from the beginning of December has been on the rise after the fall. The advantage of microalgae breeding in early succession are Cyclotella sp.,Navicula sp.,Oocystis borgei and S.quadricauda,and Chlamydomonas sp.,Chlorella sp.,S.quadricauda,Golenkinia radiata and Pseudanabeanain in medium-term, and Microcystis,Pseudanabeana,Chlorella sp. and S.quadricaudain the later period,and Microcystis,Pseudanabeana,Oscillatoria sp.and Chlorella sp. in the late,and Chlorella sp.,Golenkinia radiate,S.quadricauda in the late. The change of population succession and environmental factors closely related to the advantages of microalgae in low temperature,nitrogen and phosphorus are low and high N/P and early water salinity in diatoms and green algae;with the rise of temperature,water desalination and total nitrogen and phosphorus accumulation in algae is the dominant species of high temperature and rich;the nutrition condition of the late strong cyanobacteria became the dominant species,which has its high temperature and algae pollution resistant; late by cyanobacteria is the absolute advantage,the temperature decreased to chlorophyta was the dominant species of planktonic microalgae biomass pool decreased.

    Grape(Vitis vinifera L.) is one of the most widely used crops for plant growth regulators. In order to give retrospection about the application of plant growth regulators in grape production in China, present study used statistical method to analyze the pertinent information in this field on Chinese Pesticide Information Network. The results show that:grape on the application of plant growth regulator manufacturers are mainly distributed in the East China region such as Jiangsu,Shanghai, southwest area such as Sichuan, Chongqing and grapes important producing areas such as Shaanxi,Henan,Shandong.Among them,Sichuan province registered the largest number of 16 products; Shanghai and jiangsu have registered nine and seven products respectively. The preparation mainly took single agent as the principal, accounting for 91.80%. There are 7 kinds of registration forms, all of which are traditional formulations,mainly with solution, emulsifiable concentrates and crystalline powder, accounting for 73.76%. In the case of a single agent registration form, forchlorfenuron,thidiazuron,brassinolide,gibberellic·thidiazuron, gibberellic·forchlorfenuron,benzylamine gibberellic acid and the like are all available as a solution. Gibberellin acid has the most dosage forms,up to 6 kinds of dosage forms. The low content of gibberellic acid is mainly based on emulsifiable concentrates and soluble powders,and the high content of gibberellic acid is the crystalline powder.

    In this research,17 strains of the endophytic fungi were separated from the leaves and bark of Metasequoia glyptostroboides with different colony morphology,and the strains with antagonistic action were screened by dual culture method against plant pathogenic fungi including Fusarium proliferatum,F. graminearum,Setosphaeria turcica,Helminthosporium maydis. 1 strain was obtaind with antagonism to these strains above,named SS-17. With the morphology and ITS molecular identification of SS-17 strain,it was identified as Epicoccum nigrum.

    The experiment was conducted to screen the field pesticides against Mythimna separata(Walker),the results showed that,abamectin·chlorantraniliprole and emamectin benzoate had good control efficiency on M. separata(Walker),with high security to popularize. Otherwise,by yield monitoring,if the right medicine was chosen and sprayed in time,the corn plants with hard damaged early could recover 70 percent of the production.

    In order to enrich and change Enshi tea tree breed type, provide abundant germplasm resources for breeding of Camellia sinensis,seven varieties of tea tree including Zhongcha 102,Zhongcha 108 and Zhongcha 302 were introduced into Enshi. Because the success of tea tree introduction depends on the environment of introduction and the varieties of tea plant,as a result,this paper studies and analyzes the total annual average temperature,annual average precipitation,solar radiation and other ecological conditions in Enshi,and the tea variety adaptability,resistance,quality characteristics and phenological period,basic morphological characteristics,biochemical components,etc.,were watched and recorded, and the sensory evaluation was conducted.

    The research aimed at studying the effect of different concentrations of nanometer iron oxide on tomato growth. Using different concentration of nanometer iron oxide to deal with tomatoes,and measure the biomass,anti-oxidizing system,lipid peroxidation and contents of soluble sugar. The results showed that,after dealing with different concentration of nanometer iron oxide,the biomass had obvious change;and anti-oxidizing enzymes,contents of soluble sugar had marked variation. The trend mainly showed first increasing then falling,and tomatoes had the best growth status at 50 mg/L of nanometer iron oxide,showing the obvious growth-promoting effect. It was concluded that,different concentration of nanometer iron oxide had influence on the tomato growth,low concentrations can promote growth,while high concentrations can cause noticeable oxidative stress reaction of tomato.

    In order to elucidate the effects of harvest time on active oxygen metabolism of Feijoa sellowiana fruit during storage and determine the best harvest time,the active oxygen metabolism and AsA-GSH cycle in mitochondria of Feijoa sellowiana pulp harvested at initial ripe stage (IRS,130 d after flowering),full ripe stage (FRS,150 d after flowering) and excess ripe stage(ERS,170 d after flowering) were studied during the storage. The results showed that the H2O2 content and O2-· production rate in FRS fruits was significantly higher than those in IRS and ERS fruits during the late storage stage,but there were no significant difference before 6 d storage. At the same time,there were no significant changes in APX,DHAR,GR activities and AsA and GSH contents during the early stage of postharvest storage. After storage 9 d,the rapidly increasing trend in these indexes were showed and the biggest increasing rate was found in ERS fruits,followed by ERS fruits and FRS frruits. Therefore,in the case of active oxygen metabolism and AsA-GSH cycle during the fruit storage period,harvesting time had no significant difference on the F. sellowiana fruits in short-term storage,but for long-term storage,the full ripe stage(150 d after anthesis) was optimal harvesting time.

    In order to obtain a large number of Bletilla striata seedlings,expand the cultivated area and realize the large-scale field production,L9(33) orthogonal experiment was conducted to study the environmental conditions of B. Striata seeds.The results showed that,the combination of substrate with seedling substrate∶matured organic matter=1∶1,temperature 25 ℃,light duration 16 h/d and humidity 100% was the most favorable for the germination of B. striata seeds,with the emergence rate reaching up to 69.45%. The impact degree of temperature was the biggest,followed by matrix ratio,and the third was illumination duration.

    The drought tolerance of Malus was studied by determining leaf morphology,physiological and biochemical reactions,the aim is to provide basic data for the drought tolerance of Malus. The results showed that,drought tolerance of M. sieversii was higher than others,followed by M. zumi and M. micromalus,and the drought tolerance of M. hupehensis was the weakest among examined species.

    For the demand of saving the input in protected cultivation in South China,the effects of different pollination methods on fruit set and abortion of melon and watermelon were investigated. The high temperature tolerance between Pinghu black bee and Pinghu wasp was compared,and the effects of different placement in the bee box on Pinghu black bee and Pinghu wasp during hot season was studied. The research also worked on the fruit setting rate,fruit abortion rate,single fruit weight,quality and yields among three different pollination methods(Pinghu black bee,Pinghu wasp and human pollination) on melon and watermelon cultivation. The results showed that Pinghu black bee had better tolerance and less injured influenced by high temperature in summer. And,putting 1/10 of the beehive box into greenhouse and remaining 9/10 outside,showed 5 day length elongation of pollination period,and prolong the life span of the bee population for 30 days. Pinghu black bee showed better pollination ability among small and medium size watermelon, and muskmelon cultivation,which increased the fruit setting rate and reduced fruit abortion rate. However,the single fruit weight was lighter,when compared Pinghu black bee pollination to human pollination. The brix in the central part and edge part of fruit showed higher results,which compared Pinghu black bee pollination to human pollination method. Pinghu black bee pollination method showed the highest yields per 667 m2, which increased the economics output of muskmelon and watermelon industry.

    An acute toxicity test of Eprinomectin in situ gel injection on mice was taken to evaluate its safety by simplified Karber's method. The dosage and grouping were determined based on the minimum lethal dose(LD100) and the maximum tolerated dose(LD0) in preliminary experiment. The mice were administered with Eprinomectin in situ gel injection and observed seven days. The number of deaths was recorded for calculating the LD50(lethal dose 50%) and the 95% confidence limit. The results showed that the acute toxicity LD50 of Eprinomectin in situ gel injection on mice was 5.903 g/kg,95% confidence limit ranges was from 5.105 3~6.415 0 g/kg. It indicated that the Eprinomectin in situ gel injection was non-toxic substances according to chemically drug classification.

    In order to investigate the roles of expression of Hedgehog and NF-κB signaling pathway signal molecules and inflammatory cytokines in the alveolar epithelial typeⅡ cells (AECⅡ) infected with Mycobacterium tuberculosis(Mtb),the human AECⅡ cell line A549 was infected with Bacillus Calmette-Guérin(BCG,attenuated strain) and H37Rv (international standard virulent strain) respectively,and the signal molecules of Hedgehog and NF-κB signaling pathway and inflammatory cytokines in A549 cells of immune response to Mtb was detected by qPCR,and Western blot. The results showed that, the expression of Shh,Ptch and Gli1 in Hedgehog signaling pathway and the phosphorylation of NF-κB in NF-κB signaling pathway was increased in A549 cells when infected with H37Rv and BCG,and at the same time,the expression of IL-8 and TNF-α was also upregulated. And the protein expression of Shh,Ptch,Gli1 and NF-κB in A549 cells infected with H37Rv was higher than that infected with BCG. The Hedgehog and NF-κB signaling pathway could be activated, and the expression of inflammatory cytokines could be promoted in AECⅡ cells when infected with Mtb. The activation level of the both signaling pathway infected with Mtb virulent strain was higher than that of infected with BCG strain.

    The total polyphenols were extracted by ethanol reflux method, and the D-101 macroporous resin was used to purify the total polyphenols to study the effect of antioxidation and antibacterial activities on polyphenols from ginkgo(Ginkgo biloba L.) fruit sarcotesta separation by macroporous adsorption resin. The macroporous adsorption resin was eluted by 30%,50%,70%,90% ethanol and to inspect the polyphenol content and the antioxidant and antibacterial activity of different elution. Results showed that 90% ethanol elution had the highest polyphenol content and the highest antioxidant activity. The results showed that the antibacterial activity of 90% ethanol elution on the bacteria from carrot juice was significantly higher than that of crude extract. D-101 macroporous resin is suitable for the preliminary purification of polyphenols from ginkgo fruit sarcotesta.

    Apple(Malus pumila Mill.) peel was extracted with ultrasound assistance using ethanol aqueous solution containing NaOH as extracting solvent, the extraction conditions were optimized by response surface method. The extract was concentrated, and its pH was justified to 3.0 with hydrochloric acid. The mixture was filtrated and filter cake was washed with 10% ethanol aqueous solution. The filter cake was dried, and then degreased by petroleum ether. The residue was crystallized and recrystallized in ethanol to obtain ursolic acid crystal. This method was very simple and easy to scale up, which can make apple peel valuable and can reduce environmental pollution.

    Based on the use of Chlorella vlgaris in different environmental factors pH and salinity conditions,determination of dissolved oxygen and liquid algae growth, preliminary determination of Chlorella vlgaris has wide adaptability to salinity, growth in 0 to 45 in the salinity of sea water can, in which salinity 25 of the dissolved oxygen and the largest amount of growth to the largest data,the highest value of dissolved oxygen content and growth were 7.65 mg/L and 2.65×107/mL, Chlorella vlgaris at pH 5.5~11.5 environment can survive at pH 9 dissolved oxygen and growth of most high the highest the data here, the highest value of dissolved oxygen content and growth were 8.04 mg/L and 2.5×107/mL, the pH is greater than chlorella growth slowed significantly in 11, and even the phenomenon of death.

    The method of simultaneous determination of 4 kinds of tocopherol in vegetable oil was established by reversed-phase liquid chromatography.After sample ultrasonic extraction with methanol,centrifugal supernatant liquid nitrogen blow to dry,the capacity after the methanol organic phase filter membrane,PEF column(150×3.0 mm,2.6microns) separation,methanol and water (88∶12) elution flow equal degree,velocity 0.4 mL/min;Column temperature 30 ℃;The fluorescence detection (a) of the sample volume was 5 μL,the external standard method was quantitatively determined,and the two types of paroxygenol isomers were isolated at the same time in 20 min. The mass concentration of 4 kinds of tocopherol was in a good linear relationship with the peak area within the range of 0.5~50.0 μg/mL,and the linear correlation coefficient was greater than 0.999. Adding in the three levels with respect to the average recovery was 84.47%~116.50%,relative standard deviation(RSD) for 6.0% or less(n=6),alpha tocopherol,beta tocopherol,gamma tocopherol and delta-tocopherol,method detection limit (LOD) were respectively 0.10,0.05,0.05,0.04 mg/kg. The method is accurate,sensitive and reliable,and can be applied to the analysis of actual samples.

    Determination of selenium content in mulberry leaf tea by ultrasonic-assisted hydride generation atomic fluorescence spectrometry has the advantages of simple operation,short time period and accurate result. The detection curve equation is I=119.495 1C-0.564 1,the correlation coefficient R=0.999 8, the recovery range is 92.38%~107.13%, the average recovery rate is 98.89%. The results showed that Sanyang town mulberry leaf tea selenium content reached the highest, reaching 1.771 5 mg/kg, Guangfo town mulberry leaf tea content is the lowest,only 0.012 1 mg/kg. Sanyang Town and Xihe Town produced by the mulberry leaf tea,gingko leaf tea and Xinglong town of Eucommia leaf tea have reached the standard of selenium.

    The contents of 1-deoxynojirimycin (DNJ) in spring leaves from 24 representative cultivars of mulberry in Ankang city were determinated using reversed-phase high performance liquid for promoting the development of sericulture industry and the development of mulberry products. The results indicate that the contents of DNJ in different mulberry cultivars have significant differences. The content of DNJ in leaves of Xunyangqingjing is 0.3815%,it is significantly higher than the other varieties. The contents of DNJ in leaves of Daping No. 2 variation,Heng No. 2,Anxuan No. 1, Liushuiqingsang, Local husang,Shen No. 2,Dapingtongsang,Bolansang, Qianjinsang are relatively high; the contents of DNJ in leaves of Guangmingsang,Jingsang are relatively low. The variety of Xunyangqingjing could be as material for exaction DNJ because of the highest content of DNJ.

    The interaction between matrine (MAT) and human serum albumin (HSA) and bovine serum albumin (BSA) were investigated by fluorescence spectroscopy,UV absorption spectroscopy and circular dichroism spectroscopy with the simulated physiological condition. The experiment results of show UV absorption spectroscopy and fluorescence spectroscopy that matrine can react with HSA or BSA and form a kind of new compound at the ground state and prove that the flurescence quenching of HSA or BSA by matrine is dynamic quenching,and at different temperature,quenching constants are respectively HSA:4.60×103,1.71×104,3.07×104 L/mol and BSA:3.17×104、3.91×104 and 4.48×104 L/mol; binding constants are respectively HSA:5.999×105、3.097×106,6.667×107 L/mol and 1.007×105,1.163×105,1.582×105 L/mol, the number of binding sites is approximately equal to 1. The calculation results of thermodynamic parameters display that the reaction is hydrophobic effect and is spontaneous reaction between matrine and HSA or BSA.

    TLR4 could detect endotoxin by identifying lipopolysaccharide (LPS) and play a key role in host defense mediated by Gram-negative bacteria(LPS),therefore, TLR4 is a significant candidate gene for rabbit diseases resistance research. To identify nucleotide polymorphisms between two different breeds we designed primer pairs that amplified fragments of coding sequence(CDS) of 12 individuals, and compared the sequences. Comparative sequence analysis indicated that 3 SNPs were identified, all were non-synonymous SNPs. Two nonsynonymous SNPs(G628T and A653C) were located in the LRR domains of the predicted protein. The nonsynonymous SNPs in the LRR domains, especially related to leucine residues, may dramatically alter the ability to identified extracellular pathogens. The identification of these polymorphisms reported here increases the resource of genetic markers useful for association analyses with disease resistance and provides an important theoretical basis for disease resistant breeding of meat rabbits.

    The hyperspectral characteristics of the typical representative of wetland vegetation moss grass,reeds,south should realize,Yi grass,water smartweed, artemisia selengensis and bermudagrass of Poyang Lake south Los Angeles wetland national nature reserve,based on the analysis of spectral reflectance data, using first-order derivative,the second derivative method to analyze the reflection spectrum band characteristic diversity of vegetation types. The results show that the spectral reflectance of these seven species in the visible band is lower than 15%. In "red edge" position the absorption rate of size of artemisia selengensis>bermudagrass>carex>water smartweed reeds>the national south Yi grass,reed,red edge slope. First derivative method can't get the best characteristics of vegetation spectral bands, but the second derivative effect is very good, according to seven kinds of typical vegetation type in 685.8 nm,692.4 nm,698.6 nm,704.3 nm,737.5 nm and 746.2 nm, which have obvious difference. The establishment of typical vegetation spectral library will provide basic data for wetland vegetation classification.

    The original image is depicted and sharpen by convolution filtering, and then put it in HSV model to obtain the image color extraction. In order to get the target area of the tea wheel spot,it is also used the image binaryzation to complete the refiltering.Finally,that is a picture rapid diagnosis of tea leaf spot disease. The results showed that the image recognition of tea leaf spot in leaves of tea leaves was finished. By using convolution filtering and HSV model image processing technology can be better identified the round spot of tea lesion area, especially the clear image,size of lesion area. Taking advantage of the embeddedness of Python language,it lays the foundation for the accurate spray system of tea tea wheel spot disease.

    Based on the 2003-2014 data of financial leasing in Jiangsu province,the impact of the financial leasing industrial development on the local economic development of Jiangsu was analyzed. The results showed that,the development of financial leasing industry in Jiangsu promoted the local economy of Jiangsu province,and the amount of fixed assets investment,the number of financial leasing industry were positively correlated with the total economic output of Jiangsu province,and there was a negative correlation between the non-performing loan amount of financial leasing industry and the total economic output of Jiangsu province.Accordingly countermeasures and suggestions for promoting the development of financial leasing industry in Jiangsu were put forward.

    Based on a balanced panel data of listed securities companies from 2013 to 2015,the influences of internal governance on enterprise growth were investigated from the following five aspects:board of directors,executive incentive,ordinary staff input,the board of supervisors and ownership structure. The results showed that, among elements of corporate internal governance,there was a positive correlation between enterprise growth and the executive pay level,human capital investment and ownership concentration ratio,meanwhile,there was a negative correlation between enterprise growth and the number of board meetings,the scale of the supervisory board.

    Burning straws contributes to one of causes for the serious air pollution in China,which is caused by the change of rural production pattern and lifestyle,the thorough solution of this issue should rely on the systematic,comprehensive social and economic measures. After analyzing the command and control,the law regulation mechanism with economic incentive and public participation as well as its effectiveness of the current prohibition of burning straws in China,it was concluded that the poor implementation effect of governing measures was rooted in the environmental rights. The dual structure of urban and rural areas resulted in the weak environmental awareness,relatively low environmental demand and bad environmental governance capacity of rural residents. Thus,to obtain the desired effect,the costs and benefits of peasants must be balanced in governance.China may consider driving the policies of agricultural scale,mechanization and social ecologicalization based on the ecological compensation and in combination of national biomass energy development,strengthening the support and subsidies of national finance and social governing funds for the straw treatments,making use of various incentive measures,promoting a legal governance for solving burning straws by the economic mean based on balance of costs and benefits.
