Study on callus induction technology of Actinidia arguta leaves

Abstract: In order to solve the problem of low induction rate and poor rapid propagation effect of explants in the primary culture of Actinidia arguta (Sieb. & Zucc) Planch. ex Miq., the leaves of Actinidia arguta were used as explants, and the optimal sterilization procedure was selected by using different disinfection reagents and disinfection treatment time. Different mediums and different concentrations of KT, 2,4-D were combined to select the best initial culture medium formula. The results showed that the expanded leaves were selected as explants, treated with 75% CH3CH2OH for 30 s, then treated with 10% NaClO for 20 min, and treated with 0.1% HgCl2 for 10 min. The explants were rinsed with sterile water for 3~4 times, and then transferred to the medium MS+0.5 mg/L 2,4-D+1.0 mg/L KT+3.00% sugar+0.55% agar, pH 5.8. The medium was placed in light of 2 500 lx, 8 h/d, and cultured at 26 ℃ for 8 d. A small number of callus appeared, and the average single-bud more than 0.8 g after 60 d. The average length of single callus was 0.7~1.4 cm, belonging to medium callus.
