Comparison of RNA extraction methods from tomato leaves and fruits

Abstract: Taking tomato variety Moneymaker(MM) as the material, compared the extraction effects of three methods including kit method, Trizol method and Trizol improved method on total RNA from tomato leaves and fruits at different stages, and measured the concentration, purity and integrity of total RNA extracted samples by nucleic acid protein detector and agarose gel electrophoresis respectively. The results showed that in terms of tomato leaves, the purity of total RNA extracted by the three methods was good; Among the three methods, Trizol improved method was the most suitable method for extracting total RNA of tomato leaves. For the tomato fruits of different periods, the Huayueyang kit method had the best effect of extracting total RNA from the fruit in the green fruit stage and the color transformation stage, with the highest concentration. The cost was the highest, and the best method for extracting the mature fruit RNA was the Trizol improved method, which had the best integrity and lower cost. Functional research and other molecular biology research provide a technical basis.
