Optimization of extraction technology of effective components from upper stem and leaf of Polygonatum sibiricum by orthogonal method

Abstract: Using the aboveground stems and leaves of Polygonatum sibiricum as raw materials, the effects of solid-liquid ratio, extraction temperature, extraction time and ethanol volume fraction on the yield of flavonoids, polyphenols and polysaccharides were studied by single factor experiment. The extraction process of flavonoids, polyphenols and polysaccharides was optimized by orthogonal test. The results showed that the optimum conditions for extracting flavonoids were the extraction temperature 70 ℃, solid-liquid ratio 1∶80(m∶m), extraction time 2.5 h, ethanol volume fraction 90%, average yield of flavonoids was 4.01%. The optimum conditions for extracting polyphenols were the extraction temperature 80 ℃, material-liquid ratio 1∶40, extraction time 1.0 h, ethanol volume fraction 60%, average yield of polyphenols was 0.62%. The optimum conditions for polysaccharide extraction were the extraction temperature 80 ℃, solid-liquid ratio 1∶20, extraction time 2.0 h, and the average yield of polysaccharide was 18.61%. The test results showed that the three optimized processes were stable and feasible.
