Optimization of induction conditions for anther culture of indica rice

Abstract: Anther culture was carried out with F1 generation of Fudao 88//Huazhan/Yuyou 2640 as material. The orthogonal test method was used to set four levels of pretreatment temperature, pretreatment time, heat shock temperature and concentration of thiazolidine(TDZ); a total of 16 treatment combinations were used to induce anthers under different conditions. The anther induction rate was compared under different conditions. The results show that the optimal combination of the four factors determined by visual analysis ispretreatment temperature 10 ℃, pretreatment time 8 d, heat shock temperature 37 ℃, TDZ concentration 0.10 mg/L. However, considering the interaction between the factors, the optimal combination was obtainedpretreatment temperature 10 ℃, pretreatment time 8 d, heat shock temperature 37 ℃, TDZ concentration 0.05 mg/L.
