Cloning and bioinformatic analysis of SpNHX1 gene from Spinacia oleracea L.

Abstract: NHX1 gene was cloned from cultivated spinach (Spinacia oleracea L.) and named SpNHX1. Bioinformatics software were used to analyze the obtained gene nucleotide sequences and protein sequences encoded. The results showed that SpNHX1 belonged to the Vac subfamily, with a sequence length of 6 090 bp, an open reading frame of 1 662 bp, and 553 amino acid residues encoded. The bioinformatics analysis of SpNHX1 was carried out by using relevant software or online tools. The results showed that the molecular formula of its coding protein was C2 818H4 363N697O772S25, which was a stable hydrophobic protein. Evolutionary tree analysis shows that SpNHX1 is closely related to AtNHX1 and AtNHX2 proteins of Arabidopsis thaliana, which belong to proteins located on vacuolar membranes. SpNHX1 protein is closely related to NHX proteins of dicotyledons.
