C型产气荚膜梭菌生产用菌种种子批的建立与鉴定

Establishment and identification of seed batch of C. Perfringens type C for production

摘要: 为建立C型产气荚膜梭菌(Clostridium perfringens)生产用菌种种子批,以C型产气荚膜梭菌C59-2株第2代基础种子作为原始种子,对原始种子通过接种改良厌气肉肝汤和肉肝胃酶消化汤连续传代至第13代,分别对基础种子1代(F1)、第5代(F5)、第10代(F10)和第13代(F13)基础种子进行纯粹检验、培养特性鉴定、生化特性鉴定、产毒素能力的测定、血清型鉴定、免疫原性鉴定。结果表明,各项指标均合格,可作为仔猪大肠杆菌病、C型产气荚膜梭菌病二联灭活疫苗的生产用菌种用于疫苗的生产,基础种子批定为F1~F10代。

Abstract: In order to establish the seed batch of Clostridium perfringens type C,taking the second generation basic seeds of C. perfringens C59-2 strain as the original seeds, it was continuously subcultured to 13 generations by inoculating the appropriate medium. The basic seeds of generation 1(F1), generation 5(F5), generation 10(F10) and generation 13(F13) were carried out purity test, culture characteristics, biochemical characteristics, toxin production ability, serotype and immunogenicity identification. The test results of immunogenicity identification showed that all indexes were qualified. It could be used for the production of piglet colibacillosis and Clostridium perfringens type C inactivated vaccine. The basic seed batch was F1~F10 generation.
