含笑花花粉萌发和花粉管生长的离体培养研究

Study on pollen germination and pollen tube growth culture in vitro of Michelia figo

摘要: 以含笑花[Michelia figo (Lour.) Spreng.]花粉为材料,采用液体培养法,探讨不同浓度的硼酸(H3BO3)、钙(Ca2+)、蔗糖、聚乙二醇(PEG-4000)培养基组分以及pH对含笑花花粉离体萌发和花粉管生长的影响。结果表明,H3BO3、Ca2+、蔗糖、PEG-4000以及pH均对含笑花粉离体萌发和花粉管的生长有影响,最适H3BO3浓度为0.03 g/L,Ca2+浓度为0.07 g/L,蔗糖浓度为100 g/L,PEG-4000浓度为150 g/L,最佳pH为7.0,花粉萌发率高达81.74%,花粉管长度达424.17 μm。本次研究得出适合含笑花花粉萌发和花粉管生长离体培养的最佳培养基浓度组合,为其种质资源保存及新品种选育提供理论依据。

Abstract: To clear the influence of boric acid, calcium, sucrose, PEG-4000 and pH on pollen germination and pollen tube growth culture in vitro of Michelia figo, the pollen of Michelia figo was studied by liquid culture. The results showed that boric acid, calcium, sucrose, PEG-4000 and pH in the medium had a significant influence on pollen germination and pollen tube growth of Michelia figo. The optimal concentration of boric acid was 0.03 g/L, calcium was 0.07 g/L, sucrose was 100 g/L, PEG-4000 was 150 g/L, and pH was 7.0. The pollen germination rate was 81.74%, and the length of the pollen tube was 424.17μm. In this study, the optimal medium concentration of pollen germination and pollen tube growth in vitro culture of Michelia figo was obtained, which provided a theoretical basis for the conservation of its germplasm resources and the breeding of new varieties.
