红藜种子黄酮物质的提取及体外抗氧化活性测定

Extraction and determination of antioxidant activity in vitro of total flavonoids from Chenopodium formosanum Koidz seeds

摘要: 以台湾红藜(Chenopodium formosanum Koidz)种子为原料,采用正交试验法,探讨酶的质量分数、料液比、酶解温度和超声时间等因素对台湾红藜种子黄酮物质提取的影响,并通过分析台湾红藜种子黄酮物质对DPPH·和·OH的清除能力测定体外抗氧化活性。结果表明,提取红藜种子黄酮物质的最佳工艺条件为质量分数0.6%的纤维素酶,料液比1∶40,酶解温度60 ℃,超声时间10 min。在此条件下,黄酮物质的提取率高达27.81 mg/g。体外抗氧化性试验显示,从台湾红藜种子中提取的黄酮物质对DPPH·和·OH的清除率分别达92.91%和94.72%,具有一定的抗氧化活性。

Abstract: Taking the seeds of Chenopodium formosanum Koidz as raw materials, the orthogonal test method was used to explore the effects of enzyme mass fraction, solid-liquid ratio, enzymolysis temperature and ultrasonic time on the extraction of Chenopodium formosanum Koidz seeds, and the antioxidant activity in vitro was determined by analyzing the scavenging ability of flavonoids from Chenopodium formosanum Koidz seeds to DPPH· and ·OH. The results showed that the best technological condition for extracting flavonoids from the seeds of Chenopodium formosanum Koidz was cellulase with the mass fraction of 0.6%, the solid-liquid ratio of 1∶40, the enzymatic hydrolysis temperature of 60 ℃, and the ultrasonic time of 10 min. Under this condition, the extraction rate of flavonoids was as high as 27.81 mg/g. In vitro antioxidant test showed that the flavonoids extracted from the seeds of Chenopodium formosana Koidz had 92.91% and 94.72% scavenging activity on DPPH· and ·OH, respectively, and had certain antioxidant activity.
