湘西地区山羊泰勒虫流行病学调查及种类鉴定

Epidemiological investigation and species identification of Theileria in goats from Xiangxi Autonomous Prefecture

摘要: 为了解湖南省湘西自治州羊泰勒虫病感染情况及种类鉴定,本研究于2018年8月至2019年8月,应用PCR法对湘西自治州不同地区共204份山羊血液样品中泰勒虫属18S rRNA基因进行检测。结果发现,该地区羊泰勒虫病阳性率为81.86%,其中黑山羊和本地山羊阳性率分别为79.52%和83.47%;舍饲较散养羊群泰勒虫病感染率低,分别为75.78%和92.10%;随机挑选16个阳性样品进行测序与分析,结果发现,本研究获得的16份样品基因序列与吕氏泰勒虫(Theileria luwenshuni)中国流行株同源性达99.50%以上,遗传进化分析发现所有样品与吕氏泰勒虫分离株均属于同一分支。本研究结果表明,泰勒虫广泛流行于湘西地区羊群中,且吕氏泰勒虫为优势虫种,应加强对该病的防控。

Abstract: The present study was conducted to investigate the infection status and genetic species of Theileria in goats from XiangXi Autonomous Prefecture of Hunan Province. From August 2018 to August 2019, PCR was used to detect the 18S rRNA gene of Theileria in 204 blood samples of goats from different regions of Xiangxi Autonomous Prefecture. The results showed that the positive rate of Theileria in goats was 81.86%, and the positive rates of that from black goats and local goats were 79.52% and 83.47%, respectively. The positive rates of Theileria in goats with house feeding and raised in free range were 75.78% and 92.10%, respectively. Also, the 18S rRNA sequences from 16 positive samples were randomly selected for sequencing and analysis. The results showed that the 18S rRNA gene sequences of these samples shared more than 99.50% genetic homology with Theileria luwenshuni strains prevalent in China, and all specimens collected in this study belonged to the same branch as that of T.luwenshuni strains. The results showed that Theileria was widely prevalent in goats in Xiangxi Autonomous Prefecture of Hunan Province, and T.luwenshuni was the dominant species among them. Therefore, the prevention and control of this disease should be strengthened.
