景天三七根、茎、花、叶中总黄酮含量测定

Determination of total flavonoids in roots,stems,flowers and leaves of Sedum aizoon L.

摘要: 景天三七(Sedum aizoon L.)根、茎、花、叶样品经粉碎后以甲醇为溶剂进行索氏提取,以芦丁为对照品,NaNO2-A1(NO33-NaOH作显色剂,在优选的最大吸收波长365 nm处检测,建立总黄酮测定的紫外分光光度法并对其含量进行测定。结果表明,在浓度8~40 μg/g吸光度线性关系良好,回归方程为y=0.017 5x+0.04(R2=0.999 9),景天三七不同部位低、中、高3个浓度芦丁添加回收率在92%~94%,RSD≤5%;景天三七根、茎、花、叶中总黄酮含量分别为(29.44±1.37)、(27.96±0.87)、(9.79±0.36)和(16.16±0.50) mg/g。研究建立了一种简单、准确的景天三七不同部位总黄酮的测定方法,为景天三七不同部位总黄酮检测及有针对性地开发利用提供科学依据。

Abstract: After the root, stem, flower and leaf of Sedum aizoon L. were crushed, the soxhlet extraction was carried out with methanol as solvent. Then, the content of total flavonoids in different organs was determined by the developed ultraviolet spectrophotometry method at the optimal maximum absorption wavelength of 365 nm with rutin as reference substance and NaNO2-A1(NO33-NaOH as chromogenic agent. A good linear behaviors over the investigated concentration ranges of 8~40 μg/g were observed with the regression equation of y=0.017 5x+0.04(R2=0.999 9) for the test. The recoveries of added rutin at low, medium and high concentration levels in different organs was 92%~94%, RSD≤5%. The contents of total flavonoids in roots, stems, flower s and leaves of Sedum aizoon L. were (29.44±1.37), (27.96±0.87), (9.79±0.36) and (16.16±0.50) mg/g, respectively. This study established a simple and accurate method for the determination of total flavonoids in different organs of Sedum aizoon L., which provided a scientific basis for the detection and targeted development and utilization of the flavonoids in Sedum aizoon L.
