一株产吲哚乙酸巴基斯坦赖氨酸芽胞杆菌的快速筛选与鉴定

Rapid screening and identification of a Lysinibacillus pakistanensis strain producing indole-3-aceticacid

摘要: 采用选择性培养基,通过96孔微孔板培养和Salkowski染色法对保藏1 920株芽胞杆菌进行产吲哚乙酸(indole-3-aceticacid,IAA)能力的快速筛选,获得12株显色较为明显的芽胞杆菌。对这12株芽胞杆菌进行产IAA能力的复筛和定量分析,获得一株产IAA为(118.23±5.55) mg/L的芽胞杆菌,通过16S rDNA序列比对分析,初步确定这株芽胞杆菌为巴基斯坦赖氨酸芽胞杆菌(Lysinibacillus pakistanensis)。

Abstract: 1920 Bacillus strains were cultivated in selective media in 96-well deep well plate and rapidly screenedthrough Salkowski colorimetric method. 12 strains producing indole-3-acetic acid (IAA) were obtained, which had obvious chromatic reaction. The 12 strains were rescreened and quantitively measured. One strain could produce IAA of 118.23±5.55 mg/L which was selected. After gene sequencing and sequence alignment of 16S rDNA, the strain was identified as Lysinibacillus pakistanensis.
