牛大力叶中总多糖、总黄酮、总皂苷含量及其抗氧化活性的研究

Study on the contents of total polysaccharides,total flavonoids,total saponins and the antioxidant activity in the extracts of leaves of Millettia speciosa Champ.

摘要: 通过热水回流提取法制备牛大力叶提取物,采用显色法结合紫外分光光度法测定该提取物中总多糖、总黄酮及总皂苷含量;同时测定其对DPPH·、ABTS+·、·OH、O2-·自由基的清除能力,以评价体外抗氧化活性。结果表明,牛大力叶的总多糖含量为(1.36±0.05)%、总黄酮含量为(0.61±0.007)%、总皂苷含量为(0.15±0.013)%,对DPPH·、ABTS+·、·OH、O2-·自由基的半数抑制浓度分别为16.11、4.78、10.26、7.51 mg/mL。牛大力叶中不同类型成分的含量不同,以总多糖的含量最高;同时在体外具有较好的抗氧化活性,对ABTS+·的抗氧化作用最强。

Abstract: The extract of leaves from Millettia speciosa Champ. was prepared by hot water reflux extraction, and the contents of total polysaccharide, total flavonoids and total saponins in the extract were determined by colorimetric method combined with UV spectrophotometry. At the same time, the scavenging ability of DPPH·, ABTS+·, ·OH and O2-· free radical was determined to evaluate the antioxidant activity in vitro. The results showed that, the contents of total polysaccharide, total flavonoids and total saponins of leaves from Millettia speciosa Champ. were (1.36±0.05)%, (0.61±0.007)%, (0.15±0.013)%, respectively. The half inhibitory concentrations of DPPH·, ABTS+·, ·OH and O2-· free radical were 16.11, 4.78, 10.26 and 7.51 mg/mL. The contents of different types of components of leaves from Millettia speciosa Champ. were different, and the content of total polysaccharide was the highest. At the same time, it had good antioxidant activity in vitro, and had the strongest antioxidant effect on ABTS+· free radical.
