烟草青枯病拮抗菌的分离筛选与抑菌活性物质研究

Isolation and screening of antagonistic bacteria and its active substance against tobacco bacterial wilt

摘要: 采用平板抑菌圈法从健康烟株根围土壤中分离拮抗菌,并进行形态培养观察、生理生化试验和16S rDNA序列比较分析。结果表明,共分离得到9株拮抗菌,其中,菌株FQ-7的抑菌作用最强,抑菌圈直径达16.3 mm。将菌株FQ-7鉴定为紫边链霉菌(Streptomyces violaceolatus)。该菌为兼性好氧型,接触酶、甲基红试验阴性;能分解淀粉与酪素,不耐盐,在10 g/L时生长速度最快;能利用葡萄糖、甘露醇、蔗糖和麦芽糖,不能利用乳糖、纤维二糖、山梨醇和甜醇;菌株FQ-7所产抑菌物质为溶解于乙酸乙酯的亲脂类物质;在加热温度40~121 ℃、pH 2.0~8.0缓冲体系、8 W紫外灯照射30~120 min条件下,相对抑菌活性分别保持在88.51%~95.40%、86.52%~99.88%和85.52%~91.05%。

Abstract: The antagonistic bacteria were isolated from the rhizosphere soil of healthy tobacco plants by plate inhibition circle method, and the morphological culture observation, physiological and biochemical tests and 16S rDNA sequences were compared and analyzed. The results showed that 9 strains of antagonistic bacteria were isolated, among which strain FQ-7 had the strongest antibacterial activity, with the diameter of antibacterial zone reaching 16.3 mm. The strain FQ-7 was identified as Streptomyces violaceolatus. The strain was facultative aerobic, negative in contact enzyme and methyl red test; It could decompose starch and casein while showing tolerance to NaCl with the fastest growth rate at 10 g/L; It could use maltose, mannitol, glucose and sucrose while could not use lactose, sweet alcohol, sorbitol and cellobiose; Antibacterial substances from strain FQ-7 were subjected to lipophilic compounds dissolved in ethyl acetate; The relative inhibition activity maintained 88.51%~95.40%, 86.52%~99.88% and 85.52%~91.05% under different proceeding conditions of various temperature(40~121 ℃), pH (2.0~8.0) and UV irradiation (30~120 min UV exposure time with 8 W lamp), respectively.
