不同产地广藿香的SCoT分子鉴定

Identification of Cablin from Different Habitats by SCoT Molecular Method

摘要: 通过提取广藿香[Pogostemon cablin (Blanco)Benth.]DNA,正交设计PCR反应体系,筛选获得广藿香PCR扩增的SCoT引物,应用NTsys 2.10e软件并采用UPGMA法进行聚类分析,对分布于广州、阳江、肇庆、湛江和海南等共16个居群的广藿香样品进行分子鉴定。结果表明,SCoT引物碱基序列共扩增出167条条带,其中多态性条带有108条,多态百分率占64.7%,各品种间的相似系数分布在0.69~0.91,不同产地广藿香分别聚为一类。表明不同产地广藿香有丰富的遗传多态性,且SCoT分子标记可以作为不同产地广藿香的鉴定依据。

Abstract: : Through the extraction of cablin[Pogostemon cablin (Blanco)Benth.] DNA and orthogonal design of PCR reaction system, screened and obtained cablin SCoT primers by PCR amplification, applied the NTSYS2.10e software and used the UPGMA method for cluster analysis to identify the samples of cablin by molecular perspective method,which was distributed in 16 groups include Guangzhou city,Yangjiang city, Zhaoqing city, Zhanjiang city and Hainan province and so on. Results showed that the SCoT primer sequence amplification of 167 bands, including 108 polymorphism bands, the percentage of polymorphic was 64.7%, and the similarity coefficient between each type distribution between 0.69~0.91, while different regions cablin were respectively divided. It was indicated that cablin has abundant genetic polymorphism, and SCoT molecular marker could be used as one of the basis for the identification of cablin.
