星点设计-响应面法优化柴胡皂苷提取工艺研究

Study on optimization of extraction process of Bupleurum chinensis saponins by central composite design-response surface methodology

摘要: 在单因素试验基础上,分析提取次数对柴胡(Bupleurum chinense)皂苷类成分提取的影响。采用Central composite design(CCD)响应面法优化柴胡皂苷类成分的提取工艺,考察提取时间、提取温度、液料比对柴胡皂苷类成分提取的影响。结果表明,柴胡皂苷类成分的最佳提取工艺参数为提取温度80 ℃、提取时间60 min、液料比8∶1(mL∶g),提取2次。在此优选条件下柴胡皂苷类成分提取含量为1.80 mg/g,与预测值1.81 mg/g有良好的拟合性,提取工艺可行,预测性良好。

Abstract: On the basis of single factor experiment, the effect of extraction times on the content of saponins in Bupleurum chinense was analyzed. The extraction process of saikosaponins was optimized by response surface method of central composite design. The effects of extraction time, extraction temperature and the ratio of liquid to material on saikosaponins were investigated. The results showed that the best conditions of total saponins of Bupleurum chinense were extraction temperature 80 ℃, extraction time 60 min, liquid-to-material ratio 8∶1(mL∶g), and extraction twice. Under the optimal condition, the content of total saponins of Bupleurum chinense was 1.80 mg/g, which was well fitting with the predicted value of 1.81 mg/g. The extraction process by response surface methodology is accurate, reliable and predictable.
