枯草芽孢杆菌BZJN1发酵培养基和培养条件的优化

Optimization of fermentation mediaum and conditions for Bacillus subtilis BZJN1

摘要: 对枯草芽孢杆菌(Bacillus subtilis)BZJN1发酵培养基及培养条件进行优化。结果表明,BZJN1菌株最适发酵培养基为玉米粉25.0 g/L、鱼粉25.0 g/L、工业蛋白胨15.0 g/L、玉米浆5.0 g/L、碳酸钙1.0 g/L、硫酸镁0.3 g/L、磷酸二氢钾0.1 g/L。最适发酵培养条件为初始pH 7.0、培养温度30 ℃、接种量4%、转速220 r/min,在此条件下,发酵液芽孢数可达9.68×109 CFU/mL,相比初始基础发酵培养基及培养条件下的发酵水平提高了约2倍。

Abstract: The fermentation media and conditions for the strain of Bacillus subtilis BZJN1 had been optimized. It showed that the composition of the optimized medium was as followings: W23Corn starch 25.0 g/L, fish meal 25.0 g/L, peptone 15.0 g/L, corn steep liquor 5.0 g/L, CaCO3 1.0 g/L, MgSO4 0.3 g/L and KH2PO4 0.1 g/L. The optimized fermentation condition was as following:Starting pH 7.0, temperature 30 ℃, inoculum ratio 4%, and stirring speed 220 r/min. The spore counts of the fermentation culture might reach 9.68×109 CFU/mL under the optimized condition with tthe optimized medium, which was about 2 times higher than that in the primary medium and fermentation condition.
